Proteomic Profiling of Colon Cancer Tissues: Discovery of New Candidate Biomarkers

Colon cancer is an aggressive tumor form with a poor prognosis. This study reports a comparative proteomic analysis performed by using two-dimensional differential in-gel electrophoresis (2D-DIGE) between 26 pooled colon cancer surgical tissues and adjacent non-tumoral tissues, to identify potential target proteins correlated with carcinogenesis. The DAVID functional classification tool revealed that most of the differentially regulated proteins, acting both intracellularly and extracellularly, concur across multiple cancer steps. Synthesis and evaluation of 2-tosylamino and 2-tosyliminopyrimidine derivatives as inhibitors of some leukocyte functions

Abstract We have studied the potential anti-inflammatory effects of 20 2-tosylamino and 2-tosyliminopyrimidine new derivatives in human neutrophils. We have evaluated their interference with some leukocyte functions and 5-lipoxygenase activity. All the compounds reduced neutrophil degranulation process at concentrations in the μM range. Besides, compounds with a phenolic substitution inhibited leukotriene B 4 biosynthesis in neutrophils and decreased the cell-free 5-lipoxygenase activity. This study demonstrates that 2-tosylamino and 2-tosyliminopyrimidine derivatives can reduce the activation of neutrophil cells which may have relevance for the modulation of the inflammatory response.

Inhibition of human sPLA2 and 5-lipoxygenase activities by two neo-clerodane diterpenoids

Abstract The inhibitory effect of two neo -clerodane diterpenoids, E -isolinaridial (EI) and its methylketone derivative (EIM), isolated from Linaria saxatilis var. glutinosa , on PLA2 and other enzyme activities involved in the inflammatory process was studied. Both compounds inhibited human synovial sPLA 2 in a concentration-dependent manner with IC 50 values of 0.20 and 0.49 μM, respectively, similar to scalaradial. Besides, these compounds decreased the cell-free 5-lipoxygenase activity and A23187-induced neutrophil LTB 4 biosynthesis. Cavernolide

Abstract The inhibitory effect of cavernolide, a novel C 21 terpene lactone isolated from the sponge Fasciospongia cavernosa , on PLA 2 and other enzyme activities involved in the inflammatory process was studied. Cavernolide inhibited human synovial sPLA 2 in a concentration-dependent manner with an IC 50 value of 8.8 μM. Besides, this compound decreased in the nanomolar range the myeloperoxidase degranulation process using different stimuli. Cavernolide also inhibited TNFα, NO and PGE 2 production in intact cell experiments.
